Abstract

This study was carried out at the Tissue Culture Laboratory, ) to evaluate the effect of BA, KIN and TDZ supplemented individually and in combination with 0.4 mg/l IBA compared with the control (Growth regulators free medium) on direct shoot organogenesis of a selected promising guava cultivar in a trial to apply the tissue culture techniques as a mean of rapid clonal propagation. the obtained results confirmed that the use of 40% Clorox for 20 min had a significant effect compared to other treatments. Moreover, the percentage of contamination free shoots of guava (percentage of success) significantly increased with increasing the concentration of Clorox. During the 1st and 2nd subculture the highest number of shoots/explant was obtained on the medium supplemented with 2 mg/l BA. While, the highest mean length of shoots/explant was obtained on the medium supplemented with 1 mg/l Kin. During the 3rd and 4th subculture the highest number of shoots/explant was obtained on the medium supplemented with 2 mg/l BA while, the highest length of shoots/explant was obtained on the medium supplemented with 1 mg/l Kin. The average number of rootlets/plantlet decreased with increasing the IBA concentration. The medium supplemented with (0.5 and 1.0) mg/l IBA give the highest average root length and highest survival percentage (80%), while the lowest survival percentage (40%) was obtained with the plantlets produced on the rooting medium
supplemented with 1.5 mg/l IBA.
