Abstract

Twelve thermophilic cellulase producing fungal strains were isolated from different thermogenic habitats and identified based on their morphological characters and sequence homology of their rRNA gene sequences. All the fungal isolates were screened for their ability to produce cellulase enzymes (FPase, CMCase and β-glucosidase). Thermoascus aurantiacus MW559792 and Mycothermus thermophilus MZ723073 showed the highest cellulase activities and were chosen for the classic optimization which showed that the first has the highest cellulosic activities. To improve cellulase activities from T. aurantiacus MW559792 Taguchi method was applied, then the enzyme was partially purified and the temperature and pH characterization of the three components of the cellulase enzymes were studied. To study the application of the enzyme in bioethanol production, rice straw was pretreated by steam explosion, after which it was analyzed by the purified enzyme and a sugar solution was obtained, which was later used in the production of ethanol by fermenting it with Saccharomyces cerevisiae.
