Abstract

Dietary fibers play an important role in the maintenance of health. In addition, higher dietary fibers intake is correlated with reduced incidence of diseases and mortality. Pectin extracted from orange, pomegranate peels, nanocellulose from orange peels and anthocyanin from pomegranate have shown to possess significant antitoxic activity against CCl4 toxicity. This study aimed to evaluate the protective effect of pectin extracted from orange, pomegranate peels and cellulose hydrolyzate from orange peels and anthocyanin from pomegranate peel on albino rats induced hepatotoxic stress using CCl4. To accomplish this aim, forty male albino rats with the same ages and weights (110 ±10 g) were divided into eight groups (5 rats/group). The groups were, negative control group (NC 1), the second, third & fourth were orally administered orange pectin, pomegranate pectin and cellulose hydrolyzate from orange peel (50mg/kg) daily for two months respectively. However, the fifth, the sixth, the seventh, and the eighth groups were intraperitoneally injected (1ml/kg) CCl4 diluted with paraffin oil 1:1 (v/v) twice every week for two months. The fifth group consider as positive control fed on basal diet, while the sixth, the seventh, and the eighth groups administered orally orange pectin, pomegranate pectin and cellulose hydrolyzate from orange peel (50 mg/kg), daily for two months respectively. All groups orally administrated anthocyanin (250 mg/kg) tow times weekly for two months, as antioxidants supplemented substance except groups 1 & 5 (negative and positive (CCl4) control groups). Throughout the experiment blood samples after 15, 30 and 60 days were taken from the eye artery and collected in vacuum tubes containing clot activator to get serum, besides EDTA tubes to analyze blood picture. serum analysis was preferred to measure ALT, AST, total Protein, Albumin, total Cholesterol, Triglycerides, Sodium (Na), Potassium (K). While blood samples in EDTA tubes were used for analyzing complete blood picture. The obtained results revealed that ALT and AST levels were increased due to i.p. injection of CCl4, Meanwhile, orally administration of pectin extracted from both orange and pomegranate peels besides cellulose hydrolyzate and anthocyanin led to reback ALT and AST levels and other biomarkers to be close to normal level of control rats. This finding shows that dietary fibers play an important protective agent against toxicity induced by CCl4 and attenuate its harmful effect.
