Abstract

Lack of topical rifampicin (RIF) preparations in the global market as well as increasing chronic wound infections as a major public health problem were the leading motive for this thesis.
Bacterial biofilms are predominant features of chronic wound infections that increase resistance to the host defence system as well as to conventional antibiotics.
Chapter 1:
Development and characterization of rifampicin-loaded lipid nanocapsules
Chapter 1 dealt with the preparation of standard and essential oil-modified LNCs followed by the introduction of RIF by phase inversion method and the in-vitro characterization of the developed formulations by determining their colloidal properties, skin-staining potential, drug entrapment efficiency, drug release profile, release kinetics and stability during storage at 4 ̊C.
Chapter 2:
Microbiological assessment of rifampicin-loaded lipid nanocapsules against MRSA biofilms
<Chapter 2 focused on in-vitro characterization of the antibiofilm capabilities of the developed rifampicin-loaded standard and EO-modified LNC formulations. The microbiological study included screening of biofilm-forming bacteria among MRSA isolates and investigating antibiofilm efficacy of the developed RIF LNCs.
The results of Chapter 1 & 2 support the incorporation of EOs into LNCs for enhanced effect and provide a synergistic antibiofilm activity of combined RIF and LIN as a nanomedicine for infected chronic wound management.
