Abstract

We examined whether the cytogenetic: detection of minimal human AML cells among normal marrow might be enhanced following stimulation with the human recombinant cytokines IL-31 GM-CSF G-CSF IL-ALPHA or TNF alpha. AML blasts (E-rosette negative, after plastic adhere”ce) from 7 patients and purified normal marrow cells (CD31 CD14 and CDI5,*gative) from 7 normal donors were incubated serum-free with these molecules and cell cycle activation (thymidine uptake1assessed amlc Is of 4 patients showed considerable proliferative activity Innonstimulated cultures. Following stimulation with one or more of these growth factOrsi the DNA synthesis Of cells of patients was Increased several (10-30) folds as compared to that of purified normal mgr ow cells. To mimick in vivo sampling of marrowq artificial cell, mixture of AML-normal marrow of Opposite sex were prepared, cultured he presepce or absen ,, of these factors for various timssi and examined cytogenetically. (Y- hrom some a”alysis) among mixtures with Only 1% AML blasts from the , latter ~N patients was increased in the stimulated cultures reavailing 15-20” AML karyotypes These model studies suggest that growth factor stimulation may facilitate the detection of minimal leukemia by cytogenetic examination, eg” in the marrow of patient, ofter chemotherapy or in case of early relapse.
