Abstract

Background : This thesis is divided into six parts in addition to references and the Arabic summary : Part I: Introduction and literature review. This part involves a general introduction about cancer and different types of anticancer drugs. Part II: A sensitive LC-MS/MS method for the determination of erdafitinib in human plasma. The method greenness was investigated, and it was applied successfully on human plasma samples. Part III: A comprehensive study of the stress degradation behavior of erdafitinib. A validated stability indicating HPLC method was developed to separate erdafitinib and its stress degradation products. Part IV : A validated HPLC-UV method for the determination of erdafitinib in mouse plasma. The method was applied for the sensitive determination of erdafitinib in mouse plasma and the pharmacokinetic parameters were calculated. Part V: Micellar enhanced spectrofluorimetric method for the determination of erdafitinib. The proposed method was successfully applied to laboratory-prepared tablets. Furthermore, the high sensitivity of the method allowed its application on spiked human plasma samples Part VI: Validated LC-MS/MS method for the determination of intra and extracellular concentration of sorafenib and its N-oxide metabolite in colorectal cancer cells. The method was able to monitor the cellular uptake and entrapment of both analytes and to prove the positive effect of the ginger derived compounds on sorafenib activity.
