Abstract

Twenty nine 29 samples were collected from different pigeon markets in Dakahlia governorate from clinically infected pigeons which suffered from nervous signs and greenish diarrhea. 19 ninteen of those pigeons were euthanized and Pooled organs (brain, spleen, lung and liver) were inoculated in ECE. Positive nine collected allantoic fluid chosen samples for HA activity were examined by RT PCR using Primer sequence of 356 base pair of fusion protein gene by Nanthakumaretal., 2000 then found 8 positive sample for RT PCR and then 4 selected samples sequenced, analysed to determine the phylogenetic characterization of PPMV-1.Proteolytic cleavage sites (residues112-117) of F protein were identified as KRKKAF, KKKKRF., KKKKAF, KRKKGF forR1, R6, R7, R9. Phylogenetic analysis based on f gene revealed that four isolates were aligned in genotype VIb/2 together with other PPMV-1 from Egypt, Iran and Croatia with accession No Pigeon/ Egypt/ MN610433 R1 Dak/2015, Pigeon/ Egypt/ MN610434 R6 Dak/2014, Pigeon/ Egypt/ MN610435 R7 Dak/2015, and Pigeon/ Egypt/ MN610436 R9 Dak/2018. Genetic sequencing of these viruses revealed presence of 3 isolate had multiple basic amino acids at the cleavage site with phenyl alanine at position 117 which is typical for velogenic strain but other isolate had mono basic amino acids at the cleavage site with phenyl alanine at position 117 as non velogenic strain. In experimental study, Two hundred and fifty 250 day old commercial saso chicks were obtained from Goger, Talkha, El-Dakahlia, Egypt and kept in an isolated room. They were fed on a balanced clean ration and water. These birds were used for vaccination and challenge.
