Abstract

The present work was designed to explore the effect of some nano-materials of different sources (natural/green and chemical) silver nanoparticles (AgNPs) on the efficacy and side effects of the antineoplastic drug (Cisplatin)® in vivo and in vitro. In vitro cytotoxicity study (MTT assay): The chemical AgNPs alone showed the weakest and neglected effect, while the loaded form of the chemical AgNPs with cisplatin resulted in reduced viability of PC3 cells. 1- Hematological assessment: Intraperitoneal administration of cisplatin induced significant decrease in RBCs, WBCs and Hb levels compared to control. 2- Effect of the tested drugs on the cardiac function: A significant increase in LDH, CK, CK-MB and MDA levels were seen after treatment of rats with i.p cisplatin with a significant decrease in TAC compared to control. Green-AgNPs treated rats induced significant increase in AST, LDH, CK, CK-MB levels and MDA concentration with a significant decrease in TAC compared to control. Histopathological findings: Few glomeruli were shrunken with widen Bowman’s space. Individual hyaline cast formation and regenerating epithelial lining of the renal tubules. 3-Assessment of Nephrotoxicity: Regarding renal gene expression, i.p injection of cisplatin significantly increased the expression of Bax and Caspase-3 genes while decreased the expression of Bcl-2 gene compared to control. Injection of green-AgNPs induced a significant decrease in the expression of Bax and Caspase-3 genes while increase the expression of Bcl-2 gene compared to cisplatin group. Loaded cisplatin/green AgNPs (same dose and duration as cisplatin alone) significantly reduced the expression of Caspase-3 gene compared to cisplatin group. Histopathological findings: The glomeruli were atrophied and necrotic. Kidneys treated with Cisplatin-loaded green AgNPs showed periglomerular leukocytic infiltration, mainly polymorphs. Few glomeruli were shrunken with widen Bowman’s space. Individual hyaline cast formation and regenerating epithelial lining of the renal tubules.
