Abstract

Epilepsy is a common neurological disorder that affects people of all ages which characterized by seizures.Ellagic acid a polyphenol found in fruits, vegetables and other plant foods, is one of the well-studied phytochemicals. In this work, EA was encapsulated in calcium-alginate nanoparticles as a replacement nanosystem for oral administration of EA.. In the present study, mice were randomly assigned into ten groups after being acclimated to normal laboratory conditions for two weeks 1.group I: control group.2.group II: Ellagic acid (50mg/kg) was dissolved in freshly prepared in polyethylene glycol then given orally every other day for 33 days (17 doses). 3.group III: polyethylene glycol given orally every single day for 33 days(17 doses) 4.group IV: Ellagic acid nanoparticle (EA NP) (50 mg/kg) given orally every other day for 33 days 17 doses). 5.group V: void nanoparticle (50 mg/kg alginate) given orally every other day for 33 days (17 doses). 6.group VI: PTZ dissolved in saline then animals injected intraperitoneally (37.5 mg/kg). The four initial injections in days (1, 3, 5, 7) then they did not receive PTZ during the next 22 days and the last 3 injections were in days (29, 31, 33) and thus animals were observed for 30 min. 7.group VII: animals received PTZ and Ellagic acid concurrently. 8.group VIII: animals were received PTZ and PEG concurrently. 9.Group IX: animals received PTZ and void nanoparticle concurrently. 10.group X: animals received PTZ and EA NP concurrently. At the end of the experiment, mice had been anesthetized; blood was obtained by cardiac puncture into the heparinized tube, and centrifuged. Based on the present data, it is suggested that free ellagic acid or ellagic acid encapsulated in nanoparticles showed significant antiepileptic activity in mice.
