Abstract

The present thesis includes four basic chapters as given below:
The first chapter includes the theoretical definitions and considerations of the subject. The introductory chapter contains the overview of the radionuclides and their importance in the fields of chemistry, agriculture and diagnostic nuclear medicine, their energies, decay mode, half lives and their uses. The importance and advantages of technetium, the physical and chemical properties of molybdenum and the radioisotopes of molybdenum their half-lives, radiochemistry of technetium radioisotopes. Production of molybdenum as a parent for technetium in the reactor and cyclotron. A brief information on the transient equilibrium growth kinetics for the 99Mo/99mTc generator, types of 99Mo/99mTc generators such as Solvent extraction, sublimation, electrochemical and chromatographic generators. The previous literatures on gel generator technique and their bed materials. The labeling methods with technetium-99m were also included. Short and brief account on the quality control of the technetium-99m eluate and for determining the radiochemical yield for 99mTc-complexes.
The second chapter describes in details the experimental section including chemicals, reagents and the equipments used during the the experimental work. The procedures and solutions for the preparation of 99Mo/99mTc generator adsorbed on CeMo column matrix. The factor affecting on the radiochemical yield of 99mTc-dompridone. This section contains techniques for the evaluation of the radiochemical yield includes paper chromatography.
The third chapter deals with the data obtained and divided into two parts

First part focuses on the preparation of the 99Mo/99mTc generator, the highest distribution coefficient value of molybdenum on the CeMo gel in both Hcl and HNO3 media at pH 1, the breakthrough loading capacity of molybdenum on the CeMo gel was 0.54 m mol/g using column inside diameter 0.6 cm, amount of bed matrix 1 g and molybdenum feeding solution 3X10-2M. The preparation and performance of the 99Mo /99m Tc generator was performed as a function of inside diameter, amount of column matrix and the flow rate of the eluent. Five grams of CeMo gel was packed in 1.1 cm inside diameter was loaded with 3X10-2 M molybdenum solution of pH 1 using 0.9% saline as eluent at a flow rate 1 ml/ min., the elution yield was found to be 78 ± 5 %. The radionuclidic purity was 99.99%, the radiochemical purity of the elueted technetium was 97±2 % as pertechnetate. The Mo and Ce content in the eleuate were found to be less than 4 and 2 ppm, respectively. The pH values of the 99mTc eluate ranged from 4 to 5.
The Second part focuses on the determination of the radiochemical yield of labeling dompridone with technetium-99m. The dompridone complex was prepared by direct labeling method in which the optimum condition that gave a high radiochemical yield using paper chromatography were summarized as follow: the radiochemical yield was 96 ± 0.41%, the concentration of dompridone was 5 mg, the pH was 4, the tin (II) content was 100μg, the reaction time was 30 min and the dompridone complex was stable up to 4 h.
