Abstract from the obtained results it could be concluded that guava nodal explant could be propagated in vitro through this protocol, surface sterilization by immersion in clorox 40 % for 15 min. the best antioxidant treatments was as following: 1- Using the second node of shoot tip. 2- Collecting explants in spring season with culturing in vertical position. 3- Sealing cut ends of collected explants before culturing with molten paraffin wax. 4- Pretreatment of explants with citric acid at 100 mg /l, 150 mg /l ascorbic acid and 0.5 % PVP. 5- Adding adsorptive or antioxidant material to ms medium (citric acid at 100 mg /l +150 mg /l ascorbic acid). 6- Incubation of cultured explants on darkness for 7 days. 7- Incubation explants after culturing on 4 º C for 7 days. The MS media supplied with 1.0, 1.5 mg /l BA gave the highest average number of shoots and Adding (1.0 mg /l BA + 0.2 mg /l IBA) and (1.5 mg /l BA + 0.5 mg /l IBA) to MS media gave the best effect on average length of explant and average number of leaves. Addition of 1.5 mg /l GA3 to half strength MS media gave the average length of explants. The best rooting of shoots was in MS medium at half strength with 1.0 mg /l IBA and the rooted plants were acclamized successfully. These treatments are recommended to guava propagation by tissue culture techniques. |
