Abstract

The I/R phenomenon is one of the factors entailed in MODS etiology, possibly via
disruption in the redox status and/or triggering the inflammatory cascade in different
organs. Separate organs subjected to I/R could expose the anti-oxidative capacity of
the biological system via the excessive release of FRs and ROS.
Hence, the current study elucidated the possible protective mechanisms of
anandamide, endogenous cannabinoid, against the I/R insult. Anandamide plays an
important role in some physiological functions, such as modulation of
pain/inflammation, protection of some organs and regulation of apoptosis program.
These effects result in clinical benefits, such as regulation of inflammatory
responses, as well as modulation of redox imbalance, which are urgently required to
attenuate MODS.
The examined groups were used to test the possible protective effect of anandamide
on functional-related parameters, as well as the anti-oxidant and anti-inflammatory
markers in several organs that could be affected by gastric I/R.
The first group served as the sham control was manipulated without occulsion, the
second was the positive control one I/R was subjected to the celiac artery occlusion,
while the third presented the anandamide-pretreated group.
Anandamide intraperitonely (1.5μmol/kg) was given 45 min before I/R, which was
carried out in the anaesthetized animals by clamping the celiac artery for 30 min
followed by declamping the artery to allow for reperfusion. After 90 min animals were sacrificed and their gastric, intestinal, hepatic, renal and pancreatic tissues, as
well as serum samples, were collected for determination of biochemical parameters.
The effect of anandamide on gastric ulcer, the redox system (MDA,
GSH,SOD,CAT), and its anti-inflammatory activity (MPO,TNFα and NF-κB,
AMPK) was evaluated in the 5 organs/serum. Additionally, the functional-related
parameters specific for each of the affected organs were assessed in the collected
serum and they include the pancreatic activity of lipase and amylase, hepatic ALT
and AST, as well as the serum content of Cr and BUN.
