Abstract

The study was conducted on a total of 81 samples, 7 of them are milk samples while there are 26 tissue samples (24 tongue epithelium, 1 gum scraping or tonsil and 1 lymph nodes), and 48 sera they were collected from cattle and buffaloes showing clear signs of fever, salivation, vesicles and ulceration in the gum and tongue. All samples are from Dakahlia governorate. A trial for isolating the virus in BHK-21 cell line was performed using tongue epithelium homogenates. Two serial passages were conducted for each sample.The nine-isolated virus were identified using antigen capture ELISA, RT-PCR with the suitable primers. Another 16 tissue sample were identified using qRT-PCR and RT-PCR followed by serotyping using conventional RT-PCR for O and A serotypes and SAT2 specific real time PCR.Seven milk samples were tested for the presence of FMDV RNA in real time PCR.Forty-eight serum samples were tested for the presence of anti-FMDV antibodies using Serum neutralization test. The results revealed that:1-Nine samples were positive in cell culture isolation showing rounding, nucleus pyknosis and detachment from the plate. 2-ELISA tested nine samples, three of them had optical density more 0.1, and two of them was of O serotype and the other one was SAT2.3-The nine tissue samples that grew in BHK-21 cells were tested by RT-PCR and 5 of them gave bands at 328 bp. 4-Sixteen-tissue sample were tested by Real time PCR, only nine of them were positive and seven were negative. 5-Then FMDV universal primer in conventional PCR assay tested the nine positive samples and all the nine were positive.6-Then with O specific primers: five samples were positive and 2 with A specific primers and one SAT 2 serotype by SAT 2 specific real time PCR assay.7-All 7 animals with milk samples were in contact with infected cases, 4 of them showed no signs, three of the four have FMDV in their milk. Two animals had signs and have viral RNA in their milk. One animal had the signs a weak before sampling also had viral RNA in their milk.8-All 48-serum samples showed antibody titer against FMDV.
